Mouse Bone Marrow-Derived Macrophages (BMDMs) are a primary macrophage model obtained through in vitro induced differentiation. They are generated by extracting bone marrow cells from the femurs and tibias of mice and culturing them for approximately 7 days in the presence of specific cytokines, allowing hematopoietic progenitor cells in the bone marrow to proliferate and differentiate into mature, functional macrophages. Due to their ease of access, ability to be expanded in large quantities, consistent genetic background (allowing the use of genetically modified mice), and their capacity to mimic key characteristics of in vivo macrophages, BMDMs have become one of the most important and widely used primary cell models in fields such as immunology, inflammatory diseases, infection immunity, and oncology.
This kit is used to differentiate mouse bone marrow cells into macrophages. This kit contains four reagents: recombinant mouse M-CSF, recombinant mouse IL-4, recombinant mouse IL-13, and recombinant mouse IFN-γ. M-CSF is the essential and uniquely critical cytokine that drives bone marrow precursor cells to differentiate, proliferate, and survive along the macrophage lineage; a concentration range of 10 - 50 ng/mL is typically used. This concentration is sufficient to support cell proliferation and differentiation without over-activating the cells. Macrophages are classified into two main types: M1-type macrophages and M2-type macrophages. LPS/IFN-γ can stimulate cells to polarize towards the M1 macrophage phenotype. IL-4 and IL-13 act synergistically to drive cells to polarize towards the M2 phenotype.

Storage
Store at -85 ~ -65°C. Valid for 1 year from the date of receipt. Avoid repeated freeze-thaw cycles; centrifuge before opening the vial. It is recommended to aliquot into single-use amounts and store at -80°C.
