Linear mRNA Synthesis Solution

Circular mRNA Synthesis Solution

Key Step & Core Enzyme Material
Step 1. Template Preparation – Plasmid Linearization
In this step, the constructed plasmid is digested with restriction enzymes to generate a linearized double-stranded DNA template for subsequent mRNA synthesis.
1.1 Bsa I, GMP Grade(Cat#10661)
The restriction endonuclease Bsa I specifically recognizes and cleaves target sites in double-stranded DNA, linearizing template plasmids to prepare DNA templates for in vitro mRNA synthesis.
Recognition & Cleavage Site:
5'······GGTCTC(N)₁↓······3'
3'······CCAGAG(N)₅↑······5'
Product Features:
High specificity for its recognition sequence.
Cleavage is blocked by CpG and Dcm methylation (complete sequence overlap).
Cleavage may be affected by EcoBI methylation (complete sequence overlap).
Not affected by Dam methylation (no sequence overlap).

Figure 1. No non-specific nuclease residues test
20 U BsaI was incubated with substrate DNA at 37°C for 1 h and 16 h, and DNA band changes were compared by agarose gel electrophoresis. The results showed that there was no non-specific nuclease residue in BsaI.
1.2 BspQI, GMP Grade(Cat#10664)
The restriction endonuclease BspQI specifically recognizes and cleaves target sites in double-stranded DNA, linearizing template plasmids to prepare DNA templates for in vitro mRNA synthesis.
Recognition & Cleavage Site:
5'······GCTCTTC(N)₁↓······3'
3'······CGAGAAG(N)₄↑······5'
Product Features:
Insensitive to Dam, Dcm, and mammalian CpG methylation.
Isoschizomers include Lgu I, PciS I, and Sap I.

Figure 2. Comparison of Digestion Efficiency at Different Enzyme Amounts (0.125–4 U): 2 U Delivers Optimal Performance
In a 50 μL reaction system, 1 μg of λDNA was treated with the corresponding amount of BspQI (incubate at 50 °C for 60 min and then incubate at 80 °C for 20 min to inactivate BspQI). Then 20 μL of the reaction solution was loaded.
M: DNA marker
C: The control group without BspQI treatment
Step 2. In Vitro mRNA Synthesis
In this step, mRNA is synthesized in vitro using a DNA template catalyzed by RNA polymerase.
2.1 T7 RNA Polymerase, GMP Grade (Cat#10625 or 10629, low dsRNA)
mRNA can be synthesized at scale via in vitro transcription (IVT). The T7 promoter is one of the most efficient promoters, enabling rapid and high-yield RNA production with T7 RNA Polymerase. It is a key enzyme for manufacturing therapeutic mRNA.
Double-stranded RNA (dsRNA), a byproduct of IVT, can trigger innate immune responses and severely impact mRNA vaccine efficacy, requiring strict control during production. The engineered T7 RNA Polymerase mutant (Cat#10629) significantly reduces dsRNA content in the final transcript.
Product Features:
Significantly reduces dsRNA content(~1/100,000)
High yield: 150–250 μg per 20 μL reaction
High co-transcriptional capping efficiency, compatible with TriLink CleanCap AG and LZCap
Excellent integrity across transcripts of varying lengths

Figure 3. Excellent RNA integrity for transcripts of varying lengths synthesized by different T7 RNA polymerases.
The yields obtained with low‑dsRNA T7 RNA polymerase (T7 mutant) are comparable to wild‑type (WT) T7 RNA polymerase. The T7 mutant exhibits superior performance in reducing dsRNA by‑product while maintaining high RNA integrity, compared with WT and Supplier N*.
2.2 Pyrophosphatase, Inorganic (E.coli), GMP Grade (Cat#10620)
Adding inorganic pyrophosphatase during in vitro RNA synthesis increases RNA yield. It catalyzes the hydrolysis of inorganic pyrophosphate into orthophosphate (P₂O₇⁴⁻ + H₂O + PPase → 2HPO₄²⁻), which both removes pyrophosphate inhibition from the reaction system and provides thermodynamic driving force to promote product formation, significantly boosting yield in large-scale mRNA vaccine synthesis.
Figures 4. The relationship between RNA yield and IPPA concentration in vitro transcription.
2.3 Nucleotides (ATP/CTP/GTP/UTP)
Nucleoside triphosphates (NTPs) are applicable to various molecular biology applications.

Figures 5. HPLC purity profiles of NTPs
Step 3. Template Removal (DNase I, GMP Grade, Cat#10611)
This step removes the double-stranded DNA template after RNA transcription is complete.
Step 4. Post-Transcriptional RNA Modification: Capping & Tailing
This step modifies mRNA to produce fully functional mRNA molecules.

4.1 Post-Transcriptional Capping
Vaccinia Capping Enzyme (GMP Grade, Cat#10614): Catalyzes the addition of a Cap0 structure to the 5' end of in vitro transcribed RNA, significantly improving mRNA stability and translational capacity while protecting against exonuclease degradation.
mRNA Cap 2'-O-Methyltransferase (GMP Grade, Cat#10612): Converts Cap0 to Cap1 using S-adenosylmethionine (SAM) as a methyl donor. The resulting Cap1 structure is less readily recognized by the immune sensor RIG-I, reducing immunogenicity while enhancing mRNA stability and translation efficiency.

Figure 6. The capping efficiency almost reached 100% by the vaccinia capping system from YEASEN. Nucleic acid mass spectrometry before the cap reaction was shown in Figure 1A. Nucleic acid mass spectrometry after the cap reaction was shown in Figure 1B.
4.2 Co-transcriptional Capping
Compatible with TriLink CleanCap AG and LZCap systems for direct co-transcriptional incorporation of Cap1 structures, streamlining the mRNA synthesis workflow.
