Interleukin-18 (IL-18) is a pro-inflammatory cytokine belonging to the IL-1 superfamily, produced by macrophages and other cell types. Mature human IL-18 shares approximately 63% amino acid sequence identity with mouse and rat IL-18. In the presence of IL-12 or IL-15, it induces Th1 cells, natural killer (NK) cells, and CD4+ T cells to produce interferon-gamma (IFN-γ), thereby amplifying Type I cellular immunity. IL-18 serves as a key cytokine in the immune response against intracellular bacteria and viruses. Furthermore, IL-18 enhances the proliferation, survival, and cytotoxic activity of NK cells and macrophages by upregulating the expression of perforin, granzymes, and FasL, thereby strengthening the clearance of tumor and infected cells.
