Dendritic cells (DCs) are the most potent antigen-presenting cells in the human body, serving as a bridge between innate and adaptive immunity. However, their extremely low abundance in vivo makes direct isolation difficult, which significantly limits DC research and applications. Nevertheless, DCs can be generated through the differentiation and induction of DC precursor cells from various tissues, such as bone marrow precursors, peripheral blood monocytes, and umbilical cord blood monocytes. For human studies, the most commonly used method is inducing DCs from human peripheral blood mononuclear cells (PBMCs), as peripheral blood is the most accessible source and yields the highest number of monocytes.
This cytokine set includes recombinant human GM-CSF, IL-4, IL-6, TNF-α, and IL-1β. IL-4 inhibits the differentiation of monocytes into CD14+ macrophages, thereby inducing their differentiation into immature DCs. Without IL-4 in the culture system, monocytes will differentiate into macrophages. GM-CSF induces the differentiation of monocytes or DC precursors into DCs, while promoting DC proliferation and survival. Furthermore, GM-CSF upregulates surface antigens on DCs, enhancing their capacity for antigen capture and processing. The combined action of GM-CSF and IL-4 drives the directed differentiation of monocytes into immature DCs. The combination of TNF-α, IL-6, and IL-1β induces the complete maturation of DCs.

Before opening the vial, centrifuge it first to bring the contents to the bottom. Res uspend with sterile water and reconstitute to a concentration of 0.1–1.0 mg/mL; f urther dilution can be performed according to subsequent experiments. For long term storage, it is recommended to add a carrier protein (0.1% BSA) to the dilutio n buffer. Aliquot into single-use amounts, freeze at -80℃, and avoid repeated fre eze-thaw cycles. |
Storage Conditions
Store at -25 ~ -15℃. Valid for 1 year from the date of receipt. After reconstitution, store at 2 ~ 8℃ under aseptic conditions, valid for 7 days. Alternatively, after reconstitution, store at -85 ~ -65℃ under aseptic conditions, valid for 3 months.
