The Human Hematopoietic Stem Cell (HSC) Expansion Cytokine Set contains recombinant human Flt3-Ligand, recombinant human SCF, recombinant human TPO, and recombinant human IL-6. These four cytokines are essential key growth factors for the in vitro culture of hematopoietic stem cells (HSCs). These factors promote the proliferation, survival, and differentiation of HSCs.

Flt3-Ligand is a growth factor that regulates the proliferation of early hematopoietic cells. It primarily acts on short-term hematopoietic stem cells and multipotent progenitor cells, prompting them to rapidly enter the cell cycle for expansion. By itself, it does not stimulate the proliferation of early hematopoietic cells, but rather acts synergistically with other cytokines to induce growth and differentiation.

SCF (Stem Cell Factor) is a hematopoietic growth factor that exerts its activity by signaling through the c-Kit receptor. It can effectively inhibit the programmed cell death of HSCs in vitro. SCF itself has limited proliferative capacity, but it can "prime" the cells, bringing them into an activated state and greatly enhancing the effects of other factors (such as Flt3-L, TPO, and IL series). It is the cornerstone of almost all efficient expansion protocols. Furthermore, similar to its interaction with membrane-bound SCF in the bone marrow microenvironment, it helps HSCs maintain their undifferentiated state and homing ability.

TPO (Thrombopoietin) has a core function in maintaining the quiescence and self-renewal of long-term hematopoietic stem cells. In expansion cultures, the presence of TPO helps maintain cells in a more primitive state, reducing premature differentiation into lineage-committed progenitor cells, thereby preserving their long-term repopulation ability after transplantation. Although its primary function is to support megakaryocyte/platelet differentiation, its role at the early HSC stage is to maintain stemness.

IL-6 (Interleukin-6) has a core role as a potent co-stimulatory factor. In combination with early-acting factors (such as SCF, FLT3-L, and TPO), it activates HSCs from the quiescent state (G0 phase) into the cell cycle, thereby initiating proliferation. At the same time, by activating pathways such as Akt, it promotes cell survival and reduces apoptosis during the culture process.

Before opening, centrifuge first to bring the contents to the bottom. Reconstitute with sterile PBS to a concentration of 0.1-1.0 mg/mL; further dilution can be performed according to subsequent experiments. For long-term storage, it is recommended to add carrier protein (0.1% BSA) to the dilution buffer; aliquot into single-use amounts, store frozen at -80°C, and avoid repeated freeze-thaw cycles.
