Description

Vitronectin (VTN) is a multifunctional glycoprotein with diverse physiological roles, present in plasma and the extracellular matrix. The human VTN monomer is synthesized as a 478-amino acid (75 kDa) precursor polypeptide, including a 19-amino acid signal peptide. Vitronectin is enriched in pericytes of the central nervous system; vitronectin-deficient mice, as well as mutant mice expressing vitronectin incapable of binding integrin receptors (VtnRGE), exhibit barrier leakage. Vitronectin regulates barrier function by binding to integrin receptors on endothelial cells. It is well established that vitronectin primarily binds integrin receptors through the RGD sequence, participating in cell attachment, spreading, and migration. Recent evidence indicates that VTN has additional functions in the nervous system, as it is involved in neural differentiation, neurotrophism, and neurogenesis, as well as in regulating axon size and supporting and guiding axonal extension. Moreover, VTN has been shown to play a critical role in brain protection by reducing blood-brain barrier permeability through interactions with integrin receptors in vascular endothelial cells.

Reconstitution Method

Briefly centrifuge the vial before opening to collect the contents at the bottom. Reconstitute in ultrapure water to a concentration of 0.1-1.0 mg/mL, which may be further diluted for subsequent experiments.

Store at -25 to -15°C, valid for 1 year upon receipt. After reconstitution: store at 2-8°C for up to 7 days, or at -85 to -65°C for up to 3 months. It is recommended to aliquot the protein upon first use to avoid repeated freeze-thaw cycles.
