Description
This product is a single-tube, probe-based RT-qPCR master mix specifically formulated for one-step quantitative PCR assays using RNA templates, suitable for both singleplex and multiplex detection. Reverse transcription and qPCR are performed in the same reaction tube, simplifying workflow and minimizing the risk of contamination.
The kit incorporates a next-generation antibody-mediated hot-start Taq DNA polymerase, aptamer-modified reverse transcriptase, and a specialized buffer system optimized for one-step, fully pre-mixed RT-qPCR. It supports pre-mixing of primers and probes; users only need to add template RNA during setup, thereby reducing handling steps and reagent loss. Additionally, the formulation includes a dUTP/UDG system that remains active at room temperature, effectively mitigating the risk of aerosol-mediated carryover contamination.
The aptamer-modified reverse transcriptase ensures near-complete suppression of enzymatic activity at low or ambient temperatures while enhancing overall enzyme stability. Validation studies demonstrate that primer/probe pre-mixed reactions remain stable—showing no performance degradation—after storage at 25°C for 2 weeks or at 4°C for 4 weeks. Therefore, accelerated stability testing is recommended at temperatures such as 25°C, 16°C, and 4°C to best reflect real-world product performance.

HPIV and RSV pseudovirus templates (1 copy/μL) were tested using a 25 μL respiratory assay. The 16914ES mix premixed with primers and probes was stored at 37°C for 5 days and compared with –20°C and freshly prepared controls. The amplification curves, fluorescence signals, Ct values, and detection rates showed no noticeable changes, confirming the strong stability and sensitivity of the fully premixed 16914ES.
