Hieff NGSTM OnePot Pro DNA Library Prep Kit for IlluminaTM is a next-generation enzymatic DNA fragmentation-based library preparation kit specifically designed for the IlluminaTM high-throughput sequencing platform. Compared to traditional library prep methods, this kit employs a high-quality fragmentation enzyme, eliminating the need for laborious sonication. It simplifies the workflow by combining the fragmentation and end-repair modules into a single step, significantly reducing both time and cost. The kit delivers excellent library conversion efficiency and is suitable for various sample types, including standard plant and animal genomes, microbial genomes, and FFPE DNA. The improved version of the kit features an optimized ligase to reduce adapter self-ligation, and a novel high-fidelity polymerase to further enhance amplification uniformity and fidelity.

3. Low GC Bias

4. Superior performance

Starting with 50 ng GW-OGTM006 DNA, libraries were prepared using Yeasen #12205, supplier V*, and supplier K* kits following their manuals. Libraries were size-selected with 0.7×/0.2× dual-bead purification, and 500 ng of each was captured using the IDT xGen Pan-Cancer Panel (801,904 bp). Sequencing was performed on the Illumina PE150 platform with equal data volumes analyzed. Results demonstrated that Yeasen #12205 achieved superior coverage uniformity and reduced self-ligation artifacts compared with other suppliers.
