This kit contains recombinant mouse IL-6, mouse TGF-β1, mouse IL-23, and mouse IL-1β, and is used to induce the differentiation of mouse naïve CD4+ T cells into Th17 cells. IL-6 and TGF-β1 act synergistically to activate the STAT3 signaling pathway. This is essential for overcoming TGF-β-induced Treg differentiation and shifting towards the Th17 differentiation program. They also induce the production of the autocrine factor IL-21, which further amplifies the Th17 response.
IL-23 is added during the middle to late stages of Th17 differentiation. By activating STAT3, it maintains the expression of RORγt and the IL-23 receptor, promotes the terminal differentiation and expansion of Th17 cells, and enhances their pathogenicity. In the absence of IL-23, the resulting Th17 cells may be unstable and prone to convert into other subtypes.
IL-1β is a potent enhancer. For mouse Th17 cells, adding IL-1β can significantly improve polarization efficiency (the proportion of IL-17A+ cells), especially when using serum-free media or when polarization efficiency is suboptimal.
These cytokines constitute the "instructive" microenvironment for Th17 differentiation and are key to the success of the experiment.

Before opening, centrifuge first to bring the contents to the bottom. Reconstitute with sterile deionized water to a concentration of 0.1-1.0 mg/mL. It can be further diluted according to subsequent experiments. For long-term storage, it is recommended to add carrier protein (0.1% BSA) to the dilution buffer; aliquot into single-use amounts, store frozen at -80°C, and avoid repeated freeze-thaw cycles. |
