Hieff Canace™ Veritas Long PCR Master Mix (No Dye) is a ready-to-use 2× premixed solution containing High-Fidelity DNA Polymerase, dNTPs, and an optimized buffer system specifically designed for long fragment amplification, with an extension rate of 10 sec/kb. Its fidelity is 83-fold higher than that of Taq DNA polymerase and 9-fold higher than that of standard Pfu DNA polymerase, making it suitable for amplification of complex templates. The amplification products have blunt ends.
Hieff Canace™ Veritas Long PCR Master Mix (No Dye) offers advantages including rapid and convenient operation, high sensitivity, strong specificity, and excellent stability. It can amplify DNA fragments with GC content ranging from 20% to 80%. The mix is capable of directly amplifying crude samples such as whole blood without prior nucleic acid extraction, and is compatible with blood preserved in EDTA, citrate, or heparin, as well as dried blood spots on cards, enabling faster and more flexible workflows. Additionally, the product contains specific stabilizers that maintain enzymatic activity even after repeated freeze-thaw cycles. All components provided in this product have undergone rigorous quality control to ensure superior performance and consistent batch-to-batch reproducibility.

Using 50 ng of human genomic DNA as template, various kits were tested for amplification of fragments ranging from 1 to 40 kb, following their respective optimal protocols. 10165ES was programmed with an extension speed of 5 sec/kb for fragments ≤10 kb and 10 sec/kb for fragments >10 kb. Results demonstrate that 10164ES successfully amplifies genomic DNA fragments up to 40 kb.

2. Broad Compatibility for cDNA Amplification

cDNA was synthesized from 293 cell RNA using the Yeasen Fast Reverse Transcription Kit (Cat#11149ES). The obtained cDNA served as the template for PCR amplification of fragments with different lengths. M1: 1 kb DNA Ladder; M2: DL15000 DNA Marker. The results indicate that Yeasen 10165ES enables efficient amplification of cDNA fragments ranging from 1 kb to 14 kb, demonstrating high fidelity (83× higher than standard Taq) and excellent performance with long cDNA targets.

Fidelity was evaluated using next-generation sequencing (NGS). The results show that Yeasen 10166ES exhibits 83× higher fidelity than Taq DNA polymerase and demonstrates superior accuracy compared with other commercial suppliers.
4. High sensitivity

Serial dilutions of Arabidopsis thaliana genomic DNA, mouse genomic DNA, and plasmid DNA (1 pg–1 μg) were used as templates for PCR amplification and gel electrophoresis analysis. M: 100 bp DNA Ladder. The results show that Hieff Canace® Veritas Long PCR Master Mix exhibits high sensitivity and reliable amplification performance, enabling successful amplification even from low-input DNA templates.

(A) Freeze–thaw stability test. The product was subjected to 10, 20, and 30 freeze–thaw cycles on ice. PCR was performed using 1 ng of human genomic DNA to amplify a 500 bp fragment and 50 ng to amplify 2 kb, 10 kb, and 40 kb fragments. M: DL15000 DNA Marker. The results show that 10165ES maintains robust amplification performance even after 30 freeze–thaw cycles.
(B) Storage stability test. The product was stored for 4 weeks at 4°C and 25°C, and for 2 weeks at 37°C. PCR reactions were performed as above. M: DL15000 DNA Marker. The results demonstrate that 10165ES remains stable under prolonged storage at various temperatures.
