Unit Definition

One unit (U) is defined as the amount of enzyme required to completely convert 1 μg of supercoiled pUC19 DNA to the nicked form in a 50 μL reaction at 55 °C for 1 h.

Notes

1. The volume of enzyme added to the reaction system should not exceed 10% of the total volume, to avoid star activity caused by excess glycerol in the enzyme storage buffer.

2. The additives in the restriction enzyme storage buffer (e.g., glycerol, salts) and the contaminants in the substrate solution (e.g., salts, EDTA, or ethanol) are of the same nature; the smaller the reaction volume, the more pronounced the inhibitory effect on cleavage.
