Hifair™ colorGPS All-in-one RT-gDNA Digestion SuperMix for qPCR is a newly upgraded, one-tube, trackable reverse transcription reagent specifically designed for qPCR. This product integrates all components required for genomic DNA removal and reverse transcription into a single tube. Only the RNA template and RNase-free water need to be added to perform the reaction. The reagent exhibits excellent tolerance to sample inhibitors, highly efficient genomic DNA removal, and high-yield reverse transcription performance. Additionally, the system contains a red tracking dye that enables visual monitoring during pipetting without affecting reverse transcription or gDNA digestion performance, nor interfering with downstream qPCR experiments, thereby reducing the risk of missed samples.

11157ES features a red tracking dye for visible pipetting during reverse transcription. The resulting red cDNA turns purple when added to the blue Hieff UNICON™ ColorGPS qPCR SYBR Green Master Mix (Cat# 11188ES-11190ES), providing visual tracking throughout the entire RT-qPCR workflow.

100 ng of RNA from human, Arabidopsis, and mouse samples were used as templates for reverse transcription using Yeasen 11157ES and a leading competitor's product (Supplier N*), following their respective recommended protocols. The resulting cDNA was subjected to qPCR quantification for multiple genes. The results demonstrate that Yeasen 11157ES outperforms Supplier N*, with particularly significant advantages observed when using mouse RNA templates.

11157ES exhibits strong tolerance to major residual substances commonly found after RNA extraction, including 5% absolute ethanol, 5% isopropanol, and 1.6 mM guanidine salts.

This high tolerance ensures robust and reliable reverse transcription performance, even when RNA samples contain trace amounts of extraction reagents, thereby minimizing the need for additional purification steps.

7. Excellent Stability (Freeze-Thaw & Accelerated Stability)

(A). Reverse transcription stability:** Performance was evaluated by measuring ΔCt (Treatment – Control). The product maintained high stability (ΔCt < 0.5) after 30 freeze-thaw cycles and incubation at 37°C for 7 days.

(B). gDNA removal efficiency:** Under rigorous stress conditions—including 40 freeze-thaw cycles, 7 days at 30°C, and 28 days at 4°C and 25°C—the gDNA removal efficiency of 11157ES consistently remained >99% for 200 ng of input gDNA.

FAQ

Q1: Can this product be used with qPCR Master Mixes from other manufacturers for downstream qPCR experiments?

A1: Theoretically, yes. However, performance with third-party reagents has not been validated. If you intend to use other brands, we recommend conducting a preliminary test to evaluate performance.

Q2: Can the cDNA product be used for standard PCR experiments?

A2: No. 11157ES is a reverse transcription reagent specifically designed for qPCR and is not recommended for standard PCR amplification. If you require standard PCR, we recommend using the Hifair™ Advance Fast 1st Strand cDNA Synthesis Kit (Cat# 11149ES, Cat# 11150ES) for efficient reverse transcription.

Q3: Is the reaction program strictly required to be 37°C for 10 min and 60°C for 5 min?

A3: The recommended conditions of 37°C for 10 min and 60°C for 5 min represent the optimal parameters for gDNA removal and reverse transcription, ensuring maximum RT efficiency. If necessary, you may try an alternative protocol of 50°C for 5–15 min to determine whether the reverse transcription efficiency meets your experimental requirements.
