The Live Cell Analysis Kit contains SuperView™ 488 Caspase-3 Substrate and Ac-DEVD-CHO Caspase-3/7 Inhibitor. The SuperView™ 488 Caspase-3 Substrate provides an effective tool for detecting cell apoptosis based on Caspase-3/7 activity, suitable for fluorescence microscopy observation and flow cytometry analysis.

Compared to other fluorescent substrates or inhibitors for Caspase analysis based on FLICA (Fluorochrome-Labeled Inhibitors of Caspases), the Substrate provided in this kit detects Caspase-3/7 activity without inhibiting the apoptosis process in intact cells. The Substrate consists of a fluorescent DNA dye coupled with the Caspase-3/7 DEVD recognition sequence. Initially non-fluorescent, the Substrate crosses the cell membrane into the cytoplasm. In apoptotic cells, Caspase-3/7 cleaves the Substrate, releasing a high-affinity DNA dye. This dye migrates to the nucleus, labels the DNA, and emits bright green fluorescence. The Substrate is bifunctional: it detects Caspase-3/7 activity while simultaneously visualizing morphological changes in the nucleus during apoptosis. SuperView™ staining is compatible with formaldehyde fixation and subsequent immunostaining experiments.
