Recombinant Ten-Eleven Translocase 2.0 (TET 2.0) is an engineered Fe²⁺ and α-ketoglutarate (α-KG)-dependent dioxygenase based on TET enzyme modification and recombinant expression. Through a series of oxidation reactions, it can convert 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC), then into 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC). With subsequent treatment using a conversion reagent or enzymatic method, 5caC is further converted into uracil (U). Through PCR, uracil (U) is converted into thymine (T), enabling DNA methylation site identification with single-base resolution and high accuracy.

The product should be stored at -25°C to -15°C for 1 year.
Instructions for use
The recommended enzyme input is 30 μg per reaction*. The reaction temperature of this enzyme is 37°C. A complete reaction generally requires 2 h, and a termination step is required.

1. This enzyme should be handled on ice after thawing.
2. This enzyme is suitable for methylation enzyme-mediated conversion, with a DNA template input of 10-100 ng.
