Visible Reverse Transcription. One Tube. One Program. Just 15 Minutes.
RT-qPCR remains the gold standard for gene expression analysis, but preparing high-quality cDNA can still be frustrating. Common challenges include:
- Colorless reaction mixes make missed or duplicate pipetting difficult to detect.
- Separate genomic DNA removal and reverse transcription increase hands-on time and contamination risk.
- Residual ethanol, isopropanol, or guanidine salts from RNA extraction can inhibit reverse transcription and compromise data quality.
To address these challenges, Yeasen introduces the Hifair™ ColorGPS All-in-One RT-gDNA Digestion SuperMix (Cat. 11157ES)—a next-generation reverse transcription solution that combines genomic DNA removal, reverse transcription, and visible pipetting guidance into a single streamlined workflow.
Unlike conventional RT kits, 11157ES produces red-colored cDNA, allowing users to visually confirm every transfer step. When paired with Hieff UNICON™ ColorGPS qPCR SYBR Green Master Mix (Cat. 11188ES–11190ES), the reaction changes from red to purple, providing continuous visual confirmation throughout the entire RT-qPCR workflow. No more wondering whether a well was missed.

Why Choose Hifair™ ColorGPS?
1. One Tube. One Program. Ready in 15 Minutes.
Say goodbye to complex setups! 11157ES pre-mixes all the essential components for both gDNA removal and reverse transcription into a single tube. Simply add your RNA template and nuclease-free water, and you are ready to go. The entire reaction completes in just 15 minutes. By minimizing tube openings and pipetting steps, you significantly reduce the risk of contamination, making your workflow both simple and reliable.

2. ColorGPS™ Visual Tracking: Say Goodbye to Pipetting Anxiety
Never second-guess your pipetting again. 11157ES features a built-in red tracking dye, making your reverse transcription process fully visible. The resulting cDNA is distinctly red. When paired with Yeasen’s blue Hieff UNICON™ ColorGPS qPCR Mix (Cat# 11188ES-11190ES), the reaction mixture turns a vibrant purple. This allows for visual tracking throughout the entire RT-qPCR workflow, ensuring you never miss a sample. Best of all, the tracking dye is completely inert and does not affect your final detection performance.

3. Higher Reverse Transcription Efficiency
Don't let low yields hold back your research. In head-to-head comparisons using human, Arabidopsis, and mouse RNA templates, cDNA synthesized with 11157ES delivered lower Ct values than leading competitor products (Supplier N*). The advantage is particularly striking with mouse RNA samples, proving that 11157ES provides superior reverse transcription efficiency.

Figure 3. Comparison of Reverse Transcription Efficiency.
100 ng of RNA from human, Arabidopsis, and mouse samples were used as templates for reverse transcription using Yeasen 11157ES and a leading competitor's product (Supplier N*), following their respective recommended protocols. The resulting cDNA was subjected to qPCR quantification for multiple genes. The results demonstra
Furthermore, the tracking dye does not compromise RT performance. Comparative testing using 100 ng and 100 pg RNA inputs showed negligible differences in Ct values between the dye-containing and dye-free formulations, confirming that the dye does not affect reverse transcription efficiency.

Comparative evaluation of 11157ES with and without the tracking dye. The results demonstrate that the addition of the dye does not compromise reverse transcription efficiency across varying RNA inputs.
4. Superior gDNA Digestion for Authentic qPCR Data
Ensure your qPCR results reflect true RNA expression, not genomic contamination. Testing with 200 ng and 400 ng of human 293T cell gDNA demonstrated that 11157ES achieves a digestion efficiency of >99.8%. It effectively eliminates even high amounts of gDNA, guaranteeing that your downstream qPCR data is accurate and reliable.

5. Exceptional Tolerance to Extraction Inhibitors
Imperfect RNA extractions shouldn't ruin your experiments. 11157ES is highly resilient against common residual inhibitors. Even in the presence of 5% ethanol, 5% isopropanol, or 1.6 mM guanidine thiocyanate, the reverse transcription efficiency remains completely unaffected (ΔCt < 0.5), giving you robust performance even with challenging samples.

6. Outstanding Stability(Freeze-Thaw & Accelerated Stability)
Built to withstand the demands of a busy lab environment, 11157ES offers excellent stability under rigorous stress conditions. Whether subjected to repeated freeze-thaw cycles or accelerated storage at varying temperatures, it consistently maintains >99% gDNA removal efficiency and high RT performance (ΔCt < 0.5), ensuring batch-to-batch reliability every time.

Figure 7. Stability assessment of 11157ES under various stress conditions.
(A). Reverse transcription stability:** Performance was evaluated by measuring ΔCt (Treatment – Control). The product maintained high stability (ΔCt < 0.5) after 30 freeze-thaw cycles and incubation at 37°C for 7 days.
(B). gDNA removal efficiency:** Under rigorous stress conditions—including 40 freeze-thaw cycles, 7 days at 30°C, and 28 days at 4°C and 25°C—the gDNA removal efficiency of 11157ES consistently remained >99% for 200 ng of input gDNA.
