RNAsafe-ICE is a novel frozen tissue transition solution that helps thaw frozen tissues while protecting RNA integrity, thereby transitioning frozen tissues into a state easily processable by conventional homogenization methods for high-quality RNA extraction. Processed frozen tissues do not need to be ground into powder before homogenization, and tissues can even be further dissected prior to homogenization. Upon addition of RNAsafe-ICE solution, the tissue transforms from a hard frozen state to a soft state; at this point, the solution permeates the tissue and inactivates RNases. After treatment with RNAsafe-ICE solution, tissues can be handled at room temperature without worrying about RNA degradation. Furthermore, samples can be safely weighed, further dissected, or subjected to various experiments without affecting RNA quality. The processed tissue samples can be directly added to lysis solutions for disruption or stored long-term at -20°C.

This product is suitable for animal tissues as well as cultured cells and white blood cells. Testing has shown it is suitable for the preservation of tissues such as liver, kidney, spleen, lung, heart, thymus, and pancreas.

This product is compatible with guanidine thiocyanate-based RNA extraction reagents such as TRIeasy™ Total RNA Extraction Reagent (Cat No.10606ES60), or spin column-based RNA extraction kits.

Application Case

1. Pre-cool RNAsafe-ICE at -70°C or -80°C.

2. Sample Preservation:

A. Animal Tissues: Immerse the frozen tissue in pre-cooled RNAsafe-ICE at a ratio of 100 mg tissue to not less than 1 mL RNAlater-ICE.

Note: The thickness of the tissue block should not exceed 0.5 cm, otherwise it will affect the penetration efficiency of RNAsafe-ICE, leading to poor RNA stabilization.

B. Cultured Cells, White Blood Cells: Collect cells, wash with PBS, then add at least 10 volumes of RNAsafe-ICE. Cap the tube tightly and invert several times. Whether the precipitate is completely resuspended at this point does not affect the experiment.

3. Transfer the tissue immersed in RNAsafe-ICE to -20°C for storage. After at least 16 hours, the tissue can be homogenized to proceed with the extraction process; if not homogenized immediately, the tissue can be stably stored at -20°C for 6 months. During this process, the tissue can be removed from the solution, cut, weighed, etc. at room temperature (note that exposure time at room temperature should not exceed 30 min) and then continue to be preserved immersed in RNAsafe-ICE.

4. RNA Extraction: Remove the tissue, gently squeeze off excess RNAsafe-ICE solution with forceps, place it in lysis buffer, and homogenize immediately. If it is a cell sample, centrifuge at 10,000 g, 4°C for 1 min, discard the supernatant, and add lysis buffer.
