This kit contains recombinant mouse IL-2, IL-4, IL-21, and BAFF for the in vitro polarization of mouse B cells.
IL-2 is typically added as an enhancing component to existing polarization systems. Its most important applications include synergizing with IL-4 to enhance IgG1/IgE class switching, and with TGF-β to enhance IgA class switching. In the early stages of the B cell response, it promotes clonal expansion and prepares cells for class switching; in later stages, it may influence the fate decision between plasma cells and memory B cells.
IL-4 is the core instructive factor for IgG1 and IgE class switching. In T cell-dependent responses, IL-4 is the most critical cytokine for inducing B cells to switch from expressing IgM/IgD to expressing IgG1 and IgE. Meanwhile, it is a potent B cell co-stimulatory proliferation factor, acting synergistically with BCR or CD40 signaling.
IL-21 induces B cell differentiation into plasma cells and antibody secretion.
BAFF promotes B cell survival and maturation.

Before opening, centrifuge to bring the contents to the bottom. Reconstitute with ultrapure water to a concentration of 0.1-1.0 mg/mL. Further dilution can be performed based on subsequent experiments. |
