Hifair™ Advanced Multiplex One Step RT-qPCR Probe Mix (UDG Plus) is a reagent kit designed for multiplex quantitative PCR (qPCR) using RNA as the template. In this workflow, reverse transcription and qPCR are performed in a single reaction tube, simplifying experimental procedures and reducing the risk of contamination.

This kit employs a thermostable Reverse Transcriptase for efficient first-strand cDNA synthesis and HotStart Taq DNA Polymerase for quantitative amplification. The kit includes an optimized MP Buffer and Enzyme Mix. The buffer already contains Mg²⁺ and dNTPs, along with components that effectively suppress non-specific PCR amplification and enhance the efficiency of multiplex qPCR reactions—enabling robust multiplex amplification while maintaining primer amplification efficiency. Additionally, the dUTP/UDG system is incorporated to effectively prevent aerosol-mediated contamination.

A 10-fold serial dilution of real sample template (Ct 36–40) was tested in eight replicates to assess sensitivity across three production batches. In parallel, two-well amplification of the positive control was performed to evaluate overall amplification performance and batch consistency.

Two batches of Yeasen 16921 reagents were stored at 37°C for 3 days and compared with reagents kept under standard storage conditions. PRRSV real samples and positive controls were amplified in parallel. The Ct values of both target and positive control remained consistent across conditions, and all samples were successfully detected.
