Description
HieffTM LongSeq Plasmid Fragmentation and Ligation Module is a dedicated enzymatic module developed for third-generation Nanopore sequencing platforms. When used in combination with Cat. No. 13304ES, it enables streamlined plasmid library construction.
This kit is optimized specifically for plasmid DNA library preparation. Following enzymatic fragmentation, end repair, and dA-tailing, no purification step is required—samples can proceed directly to barcode ligation, significantly simplifying the workflow.
Features
- Wide input range: 50 ng – 1 µg plasmid DNA
- Fast workflow: ~2 hours from DNA to library
- Enzyme-based fragmentation: controlled, low-damage digestion
- Long-read friendly: preserves plasmid integrity and long inserts
- Consistent performance: strict batch-to-batch quality control

Figure 1. High-Quality Nanopore Sequencing of Plasmid Libraries Prepared Using Yeasen Enzymatic Fragmentation(13305ES+13304ES).
Plasmid libraries were prepared from 100–200 ng input DNA through a rapid 2-minute enzymatic fragmentation process. The resulting libraries achieved an N50 of 2–5 kb, showcasing robust and reproducible long-read sequencing performance on Oxford Nanopore platforms.
