This product is a phage T7 RNA polymerase derived from recombinant expression in E. coli. It is an optimized mutant of wild-type T7, formulated as a mixture of T7 RNA polymerase variants that significantly reduce dsRNA content. Using double-stranded DNA containing the T7 promoter sequence as a template and NTPs as substrates, it synthesizes RNA complementary to the strand downstream of the promoter. Double-stranded linear blunt-ended or 5'-overhanging DNA can serve as substrate templates for T7 RNA polymerase; therefore, linearized plasmids and PCR products can both be used as templates for in vitro RNA synthesis. Modified nucleotides can be added to the substrates during transcription to prepare biotin- or dye-labeled RNA.
