NTC (no template control) qPCR using Yeasen one-tube premix (Cat#16913ES) and conventional reagents was tested on E. coli, P. aeruginosa, MecA, and human DNA systems. No amplification was detected in 48-well NTCs with Yeasen premix, confirming ultra-low background bacterial and human DNA contamination.

Seven gradient samples (1–10⁶ copies/μL) were tested using both standard (66 min) and rapid (30 min) protocols on ABI Q5. The rapid protocol showed >99% efficiency, ΔCt within ±0.5, fluorescence deviation within ±10%, and typical S-shaped amplification curves.

The reagent was tested under accelerated conditions (37°C for 5 days) and refrigerated storage (4°C for 14 days), with performance compared to freshly prepared controls(-20℃). Consistent amplification curves, Ct values, and detection rates were observed across human gene, pathogen detection targets, demonstrating high stability and sensitivity.
