This product is a single-tube, probe-based RT-qPCR master mix specifically designed for one-step quantitative PCR assays using RNA templates, suitable for both singleplex and multiplex detection. During the assay, reverse transcription and quantitative PCR are performed in the same reaction tube, simplifying workflow and minimizing the risk of contamination.

The kit utilizes a next-generation antibody-mediated hot-start enzyme, a one-step optimized reverse transcriptase, and a specialized buffer system tailored for fully pre-mixed, single-tube RT-qPCR. It supports pre-mixing of primers and probes; users only need to add the RNA template at the time of setup, effectively reducing handling steps and reagent loss. Additionally, the formulation includes a dUTP/UDG system that remains active at room temperature, significantly mitigating the risk of aerosol-mediated carryover contamination.

Fully premixed multiplex primers and probes combined with Yeasen Cat#16913 were incubated at 37°C for 5 days and 25°C for 12 days. The –20°C mix served as a reference. Performance remained consistent across all tested conditions, indicating robust stability suitable for long-term premixed storage.

HPIV and RSV pseudovirus templates (1 copy/μL) were tested using a 25 μL respiratory assay. The 16913ES mix premixed with primers and probes was stored at 37 °C for 5 days and compared with –20 °C and freshly prepared controls. The amplification curves, fluorescence signals, Ct values, and detection rates showed no noticeable changes, confirming the strong stability and sensitivity of the fully premixed 16913ES.

Yeasen’s one-step RT-qPCR mix shows higher amplification efficiency and improved overall performance compared with Supplier A’s reagent when run under a fast cycling program.
