The CpG cytosines on the sequence of Hieff™ Methylated PUC19 DNA are fully methylated, which can be used as a known "positive" standard to evaluate the efficiency and accuracy of the experimental workflow. After sequencing, theoretically the C on CpG will not be converted. By calculating how many C have been converted to T, the over-conversion efficiency of bisulfite in this experiment can be evaluated, ensuring the reliability of subsequent analysis. The full length of the PUC19 DNA sequence is 2686 bp.
