The Hieff™ Fast Cell Direct Probe RT-qPCR Kit is designed for quantitative gene expression analysis of various animal cells, including adherent and suspension cell lines, primary cultured cells, various stem cells, and iPS cells.

Eliminating the need for RNA extraction, this kit allows for direct qPCR analysis. It features a simple, low-error workflow that efficiently completes all stepsfrom template preparation and reverse transcription to gene expression analysisin as little as 1.5 hours. The kit includes both reverse transcription and quantitative detection reagents, facilitating easy and rapid gene expression analysis.

Storage

Store unopened kits (11173-A and 11173-B) at -25 ~ -15°C. Once thawed, store at 4°C and take precautions to prevent contamination. For long-term storage, keep all other components at -25 ~ -15°C. Valid for 1 year.

Gene expression analysis using 293 cells in different states as templates. Samples containing 10³, 10⁴, 10⁵, and 10⁶ cells were detected using Yeasen 11173ES and supplier T* respectively. The results demonstrate that Yeasen 11173ES consistently yields lower Ct values than supplier T* across all cell input amounts, indicating superior sensitivity and better reproducibility.

Using 293 cells as the template, gene expression levels were detected by both direct lysis and conventional RNA extraction workflows with the reverse transcription and qPCR components included in the kit. The results demonstrated that the direct amplification workflow of 11173ES delivers performance comparable to traditional time-consuming RNA extraction methods, while offering a faster and more efficient solution.

(a) Reverse Transcription (RT) System: The 11173ES RT system tolerated up to 55% cell lysis product, demonstrating robust resistance to inhibition.

(b) qPCR System: The 11173ES qPCR system tolerated up to 46% cDNA input, maintaining efficient amplification even in the presence of high concentrations of lysis-derived cDNA.

4. Excellent Stability

Stability was assessed by incubating the fully pre-mixed qPCR system (containing cDNA, primers, probes, and H2O) at various temperatures for up to 48 hours. The consistent Ct values indicate that the 11173ES qPCR system is highly stable, offering a robust solution that accommodates flexible experimental schedules and operational needs.
