Description
Dendritic Cells (DCs) are the most effective antigen-presenting cells in the human body, serving as a bridge between innate immunity and adaptive immunity. However, their scarcity in vivo and the difficulty in direct acquisition greatly limit DC research and applications. Nevertheless, DCs can differentiate and be induced from DC precursor cells in various tissues, such as precursor cells in bone marrow fluid, and monocytes in peripheral blood and umbilical cord blood. For humans, since peripheral blood collection is the most convenient and yields the highest number of monocytes, the most common method is to induce DCs from human peripheral blood mononuclear cells (PBMCs). Currently, the most widely used method for generating human DCs is the induction of peripheral blood monocytes using a combination of GM-CSF and IL-4.
This cytokine set contains recombinant Human IL-4 and recombinant Human GM-CSF. These two cytokines are key components for culturing human dendritic cells. IL-4 can inhibit the differentiation of monocytes into CD14+ macrophages, thereby inducing their differentiation into immature DCs. Without IL-4 in the culture system, monocytes will differentiate into macrophages. GM-CSF can induce monocytes or DC precursor cells to differentiate into DCs and promote DC proliferation and survival. GM-CSF can also upregulate the expression of surface antigens on DCs, enhancing their antigen capture and processing capabilities. Human IL-4 acts synergistically with GM-CSF to jointly drive the differentiation process. Typically, in the presence of recombinant IL-4 and GM-CSF, monocytes can differentiate into immature dendritic cells, which can achieve full maturation upon further stimulation.

Before opening, centrifuge first to bring the contents to the bottom. Reconstitute with sterile water to a concentration of 0.1-1.0 mg/mL; further dilution can be performed according to subsequent experiments. For long-term storage, it is recommended to add carrier protein (0.1% BSA) to the dilution buffer; aliquot into single-use amounts, store frozen at -80°C, and avoid repeated freeze-thaw cycles. |
