Description

Exosomes are small extracellular vesicles (30–150 nm in diameter) secreted by living cells, characterized by a typical lipid bilayer structure. They are present in cell culture supernatants, serum, plasma, saliva, urine, amniotic fluid, and other biological fluids. Exosomes carry a variety of proteins, lipids, DNA, and RNA, playing critical roles in intercellular communication and material transfer. They also hold great promise as early diagnostic biomarkers for various diseases.

This product is a specialized serum-free medium developed specifically for exosome production. Its main components include HEPES, L-Glutamine, Pharmaceutical-grade Albumin, Hypoxanthine, Thymidine, Phenol Red, and Glucose. It is xeno-free, serum-free, and free of growth factors. Cells can be maintained in a fully supportive state without the addition of serum, facilitating high-quality exosome harvest.

This product should be stored at 2~8℃ for 1 year. Protect from light.

Application

Exosome Cultivation: Tumor Cell Lines & T Cells

Instructions

1. Cell recovery and passaging: After thawing, culture cells in complete medium for 2–3 passages until normal growth is confirmed.

2. Stepwise adaptation – Step 1: When cells reach ~50% confluency, remove the original complete medium and replace with a mixture of 50% exosome-specific serum-free medium + 50% complete medium.

3. Stepwise adaptation – Step 2: Once cells in Step 1 reach 70–80% confluency, remove the 50:50 medium mixture.

4. Wash cells 2–3 times with 1× PBS.

5. Add exosome-specific serum-free medium and continue incubation for 24–48 hours. Collect the cell culture supernatant when confluency reaches 90–100%. This supernatant is suitable for downstream exosome isolation.
