Description
YeasenBio's insect cell transfection reagent is a high-performance transfection tool developed based on innovative polymer technology, specially designed for insect cell experimental systems. The reagent can efficiently encapsulate and protect DNA molecules (including linear baculovirus DNA, plasmid DNA) through a unique cationic polymer composite structure, significantly improving the efficiency of nucleic acid delivery, while avoiding the cytotoxicity problems that may be caused by traditional calcium phosphate or liposome transfection.
This product has excellent broad-spectrum adaptability, and can achieve stable and highly reproducible transfection effects in mainstream insect cell lines such as Sf9, Sf21 and High Five™, with a transfection efficiency of 85%-95%, and has no significant effect on cell viability and subsequent protein expression. Its breakthrough formula supports serum-containing and serum-free dual culture systems, without the need for cumbersome culture medium replacement steps, significantly simplifying the experimental process. Whether it is used for rapid construction of recombinant baculovirus, large-scale production of recombinant proteins, or conducting gene function research or virus-like particle (VLP) preparation, it can achieve efficient gene delivery within 24-48 hours, and the duration of exogenous gene expression can reach more than 72 hours.
Features
- High transfection efficiency: Transfection efficiency is as high as 90%, which is better than imported brand T
- High cell compatibility: It can efficiently transfect a variety of insect cells such as Sf9, Sf21 and High Five™
- Wide application range: Both baculovirus DNA and plasmid DNA can be used

Figures
High transfection efficiency

Figure 1. GFP expression profile under fluorescence microscope
Using 40810ES-Cell Insect reagent to transfect baculovirus genomic DNA. Transfection in 6-well plates.
Fluorescence microscope observation: 2 days after transfection, the GFP expression intensity was significantly higher than that of the control group, the cells were in good condition, there was no obvious toxicity, and the positive rate was significantly better than other suppliers.
Reporter gene quantitative data: Luciferase reporter gene detection shows that the transfection efficiency of Yisheng-Cell Insect is 2-3 times that of competing products (48-hour data)

Figure 2. Luciferase reporter gene detection of luciferase expression
The insect cell line Sf9 was transfected with 0.1 µg of luciferase expression plasmids expressing two different proteins in 96-well plates. Luciferase expression was measured 48 hours after transfection.
Protein expression detection: achieving higher infection efficiency and protein expression

Figure 3. Protein expression detection
Different brands of transfection reagents were used to transfect Sf9 (A) and High Five cells (B) to express E2 protein, and the protein expression was detected on the 5th day.
Western blot verification

Figure 4. WB verification of protein expression after plasmid transfection
Sf9 cells were transfected with 2µg of GFP expression plasmid in 6-well plates, and the expression of the target protein (GFP-6XHis) was verified by Western blot in total soluble cell lysate prepared from cells 72 hours after transfection. The result showed that target protein (GFP-6XHis) was highly expressed and the bands were clear.
