Carboxypeptidase B (also known as peptidyl-L-lysine hydrolase, arginase carboxypeptidase, EC 3.4.17.2) specifically hydrolyzes C-terminal basic amino acids of proteins, such as lysine (Lys, K), arginine (Arg, R), and histidine (His, H). It is widely used in the precise post-processing of biological products such as recombinant insulin and glucagon-like peptide-1 (GLP-1) analogs.
This kit applies the double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) principle for the detection of carboxypeptidase B residue. The carboxypeptidase B standard (20374-B) and test samples are added to the pre-coated anti-carboxypeptidase B antibody plate (20374-A), followed by the addition of diluted biotin-labeled carboxypeptidase B detection antibody (20374-C), and finally Streptavidin-HRP (SA-HRP) (20374-D), forming an antibody + antigen + antibody-Biotin + SA-HRP complex. After plate washing, TMB chromogenic solution (20374-H) is added for color development. TMB is catalyzed by HRP enzyme from colorless to blue and finally converted to yellow under the action of stop solution (20374-I). The intensity of yellow color is positively correlated with the content of carboxypeptidase B detected in the sample. This kit can be used to determine trace residues of recombinant carboxypeptidase B in samples from recombinant protein drug or fusion expression processes using carboxypeptidase B for enzymatic cleavage.
