The Hieff™ Pro Soil/Stool DNA Kit is designed to extract high-purity DNA from 250 to 500 mg of soil samples. Utilizing superparamagnetic bead purification technology, this method eliminates the need for toxic phenol-chloroform extraction, offering a simple and convenient workflow. The extracted DNA is ready for direct use in downstream applications such as PCR, qPCR, microarray analysis, and Sanger or Next-Generation Sequencing (NGS). When paired with automated magnetic bead extraction instruments, the kit enables high-throughput nucleic acid extraction.

Case Study 1: High-Humic-Acid Black Soil Samples

Compared with PBS treatment, the proprietary precipitation solution:

  • Increased DNA yield by approximately 3-fold 
  • Significantly reduced humic acid contamination
  • Improved A260/230 and A260/280 purity ratios
  • Produced cleaner DNA suitable for downstream amplification

Case Study 2: High-Humic-Acid Sludge Samples

The 18527ES kit demonstrated:

  • Higher DNA recovery
  • Strong and intact electrophoretic bands
  • Minimal DNA degradation
  • Superior removal of inhibitory substances

Agarose gel analysis showed bright, well-defined genomic DNA bands with minimal smearing, indicating excellent DNA integrity and extraction efficiency.

Case Study 3: Human Fecal Samples

Human fecal samples (200 mg input) were processed using the Yeasen Enhanced Soil DNA Extraction Kit (18527ES) on the Yeasen AP-48 Automated Nucleic Acid Extraction System and compared with a leading supplier's column-based manual extraction kit (Supplier T). All samples were eluted in 80 μL.

Case Study 4: Water Filter Membrane Samples

Water filter membrane samples were extracted using the 18527ES kit and compared with Supplier M and Supplier J using the same automated extraction platform and elution conditions.

Results

The 18527ES kit achieved DNA yields comparable to both competing products, demonstrating reliable recovery of microbial DNA from low-biomass environmental water samples.

Case Study 5: Biogas Residue and Straw Compost Samples

Biogas digestate and straw compost samples were processed using the 18527ES kit alongside Supplier M and Supplier J under identical extraction conditions.

Results

The 18527ES kit produced DNA yields comparable to Supplier J and consistently higher than Supplier M, highlighting its strong capability for extracting DNA from organic matter-rich agricultural samples.
