Description
Tko RNA Helicase is derived from the TK0306 gene of the thermophilic archaeon Thermococcus kodakaraensis (strain KOD1) and belongs to the DEAD-box RNA helicase family. This enzyme is ATP-dependent and specifically unwinds single-stranded stem-loop RNA structures. It exhibits weak activity towards double-stranded RNA (dsRNA) and single-stranded DNA (ssDNA) substrates and does not act on dsRNA substrates with 5' or 3' overhangs. With an optimal temperature of 50°C, this enzyme serves as a useful tool for specifically unwinding RNA stem-loop structures under mild temperature conditions, making it suitable for applications such as RNA structural analysis and post-transcriptional regulation research.

2. Incubate at 50°C for 30 min.

1. This product is for research use only.

3. The reaction buffer for this enzyme requires 1–2 mM ATP or dATP and 2 mM Mg²⁺. The 10×Tko Reaction Buffer already contains ATP and Mg²⁺.
4. For different substrates, the enzyme amount should be adjusted; gradient testing is recommended.
