Description
HifairTM Superpro Ⅱ One Step RT-qPCR Master Mix is a one-tube premixed kit for single or multiple quantitative PCR reactions using RNA as a template. During the experiment, reverse transcription and quantitative PCR were carried out in the same reaction tube, which simplified the experimental operation and reduced the risk of pollution. The kit supports primer and probe pre-mixing, allowing for a streamlined workflow—simply add the template to the prepared reaction mix and proceed directly to on-machine amplification.

Multiplex detection of SARS-CoV-2, Influenza A, Influenza B, and internal control (5000 copies/mL and 500 copies/mL) using Yeasen's fully premixed RT-qPCR reagent (11899ES) and supplier T*.
Yeasen's reagent successfully supported at least four-target multiplex amplification without compromising performance at low template concentrations. Results were comparable to the T brand, with slightly earlier Ct values in some channels, indicating a sensitivity advantage.
3. Sensitivity

RT-qPCR targeting Influenza A and Influenza B was performed using the HifairTM Superpro Ⅱ One Step RT-qPCR Master Mix. The reagent successfully detected all low-concentration Influenza A and B templates (8/8), demonstrating excellent sensitivity.
4. High GC-content tolerance

Comparison of Yeasen's fully premixed RT-qPCR reagent(11899ES) and traditional reagent (13650) in amplifying 1 µg of 293 RNA templates with 30–60% GC content.
Results show no significant difference between the two reagents across templates with varying GC content, indicating broad GC tolerance of Yeasen’s fully premixed formulation.
5. Ultra-high stability

Yeasen's premixed reagent showed no significant Ct shift when amplifying Influenza A and B templates at 2000 and 200 copies/mL, indicating stable performance. After 5 days at 37 °C, Yeasen (Cat#11899ES) also demonstrated lower Ct values and higher fluorescence signals compared to the T brand, confirming its superior stability.
