Firefly luciferase is a protein with a molecular weight of approximately 61 kDa. In the presence of ATP, magnesium ions, and oxygen, it catalyzes the oxidation of luciferin to oxyluciferin. During the oxidation of luciferin, bioluminescence at a wavelength of approximately 560 nm is emitted, which can be measured using a luminometer.

The Firefly Glo Luciferase Reporter Gene Assay Kit is a glow-type firefly luciferase reporter gene assay kit featuring high sensitivity and stable luminescence signal, which can meet the requirements of high-throughput detection of luciferase expression in mammalian cells.
Compared with the flash-type Firefly Luciferase Reporter Gene Assay Kit (Cat#.11401), this product has the following advantages: 1) More stable luminescence signal, providing greater flexibility for experimental design. 2) Adopting an add-mix-read operation method, no need for auto-injectors, and no need to discard culture medium, centrifuge, or perform other steps, greatly simplifying the experimental workflow. 3) No obvious irritating odor.
This kit is compatible with various commonly used cell culture media: RPMI 1640, DMEM, MEM-alpha, F12, DMEM/F12, etc., with a half-life of approximately 2 hours (22°C), meeting the needs of most high-throughput experiments.

Storage

Transported on dry ice. Store the unopened kit at -20 °C , valid for 1 year. The dissolved and aliquoted glow-type firefly luciferase substrate should be stored at -70 °C protected from light for 1 year, or at -20 °C for no more than 1 month.

No. An instrument with a bioluminescence module and full-wavelength detection (350-700 nm) is required, such as GloMax 20/20 (single-tube detection) or Biotex HTX multi-functional microplate reader (equipped with auto-injector).
It can be stored at -80°C, similar to protein storage methods. Lysed samples can be stored at -80°C for up to six months, or at -20°C for one month.
Dilute the lysed sample, or reduce the cell incubation time after transfection experiments.
