Isothermal nucleic acid amplification technology can be categorized into many types based on different amplification principles. The most common and widely applied method is the Loop-Mediated Isothermal Amplification (LAMP) technology developed by Japanese scholars including Notomi in 2000. The technical feature of LAMP is the design of four primers targeting six regions on the target gene, utilizing a strand-displacing type Bst DNA polymerase to conduct the amplification reaction at a constant temperature of 60~65°C. This achieves a 109-1010-fold amplification within 15-60 minutes.
The Bst DNA polymerase is the core enzyme in the LAMP isothermal amplification technology. It possesses strong thermal stability, strand displacement activity, and polymerase activity, enabling rapid, efficient, and specific amplification of the template under constant temperature conditions. Unlike PCR, which requires thermal denaturation cycles for amplification, the entire LAMP experiment can be completed within one hour without the need for such cycles.

01 Fluorescent Dye Method Application

In the RT-LAMP fluorescent dye amplification system, the amplification performance of the lyophilized powder cake 14405ES is consistent with that of the liquid Bst enzyme (including glycerol and glycerol-free), and both can detect 100% at a sensitivity of 50 copies/T. The specificity is good, with no detection in 48 wells of NTC (Negative Template Control).
2ăGood thermal accelerated stability
 In the RT-LAMP fluorescent dye amplification system, the lyophilized powder was subjected to storage treatments at 45°C for 7 days, 45°C for 15 days, and room temperature for 19 days, respectively. After thermal acceleration, the product performance remained consistent with that of the liquid version of glycerol-free Bst enzyme, and no peak was observed in the NTC (Negative Template Control).
3ăSupports refreezing after acceleration
In the RT-LAMP fluorescent dye amplification system, the lyophilized powder was subjected to storage treatments at 45°C for 7 days and 45°C for 15 days, respectively. The lyophilized powder after thermal acceleration treatment was then subjected to a second round of lyophilization (including single enzyme and RT-LAMP reagents). Experiments show that after the accelerated treatment and the second lyophilization, the product performance remains consistent with that of the liquid version of glycerol-free Bst enzyme, and no peak was observed in the NTC (Negative Template Control). The productâs stability is good, supporting secondary lyophilization and acceleration after secondary lyophilization.
02 pH Colorimetric Method Application

In the RT-LAMP pH colorimetric amplification system, the amplification sensitivity of the lyophilized powder cake 14405ES can reach 50 copies/T (100% detection rate), and the specificity is good, with no detection in 24 wells of NTC (Negative Template Control).
2ăGood thermal accelerated stability
In the RT-LAMP pH colorimetric amplification system, the thermal accelerated stability of the lyophilized powder 14405ES was investigated at 45°C for 15 days, the stability after secondary lyophilization following acceleration (rehydration after acceleration at 45°C for 7 days), and the storage stability after rehydration (stored at -20°C for 7 days after rehydration). The experiments indicate that the 14405ES product has good stability, with no impact on product performance after thermal acceleration, and supports secondary lyophilization after acceleration.
