Description

Firefly luciferase is a protein with a molecular weight of about 61 kDa, which can catalyze the oxidation of luciferin to oxyluciferin in the presence of ATP, magnesium ions and oxygen. It emits bioluminescence with a wavelength of around 560 nm. Renilla luciferase is a protein with a molecular weight of about 36 kDa. In the presence of oxygen, it can catalyze the oxidation of coelenterazine to coelenteramide. During the oxidation process, it emits a wavelength of 480 nm. about bioluminescence. Both bioluminescence can be measured by chemiluminescence. The detection principle is shown in the figure1:

Dual Glo Luciferase Reporter Gene Assay Kit is a glow-type dual luciferase reporter gene detection kit with high sensitivity and stable luminescence signal. This kit contains high-purity firefly luciferin and coelenterazine. In the same sample, firefly luciferase is used as a substrate to detect firefly luciferase, and then the fluorescence signal of firefly luciferase is quenched, and at the same time coelenterazine is used as a substrate to detect Renilla luciferase to achieve dual luciferase reporter gene detection. Renilla luciferase is used as an internal reference, which eliminates the influence caused by the number of cells between the wells and the transfection efficiency, which makes the detection results more accurate.

Compared with the flash-type Dual Luciferase Reporter Gene Assay Kit (Cat#11402), this product has the following advantages: 1) The luminescence signal is more stable, providing greater flexibility for experimental design. 2) It is more in line with high-throughput detection, does not need to rely on automatic samplers, and does not need to discard the culture medium, centrifugation and other steps, which greatly simplifies the experimental process. 3) No obvious irritating odor. This kit can be used in a variety of common cell culture media: RPMI 1640, DMEM, MEM-α, F12, DMEM/F12, etc. Its half-life is about 2h (22°C), which meets the needs of most high-throughput experiments.
