Streptolysin O (SLO) is a pore-forming exotoxin secreted by pyogenic streptococci, such as Group A Streptococcus (GAS). As one of the most critical virulence factors of Streptococcus, it serves as a key protein molecule in both basic research and clinical diagnostics. The "O" in its name stands for "Oxygen labile," indicating its sensitivity to oxygen. Under aerobic conditions, the toxin is oxidized and loses its activity; however, its activity is restored or maintained in the presence of reducing agents (such as cysteine) or under anaerobic conditions.
The target of SLO is membrane cholesterol. Its ability to bind to cholesterol-containing membranes is a prerequisite for its pathogenicity. After SLO monomers bind to the membrane, they aggregate to form large ring-shaped oligomers (with pore diameters reaching 25–30 nm), creating pores in the cell membrane that lead to the leakage of cellular contents and cell lysis. It not only lyses red blood cells but also kills white blood cells, platelets, and various tissue cells (such as endothelial and epithelial cells).
Furthermore, SLO assists bacteria in resisting phagocytosis. At sub-lytic concentrations (levels insufficient to kill immune cells immediately), SLO can interfere with immune cell signaling pathways, induce apoptosis, or inhibit the bactericidal function of neutrophils, thereby aiding bacterial colonization and dissemination within the host.

Storage
Store at -25~-15°C, valid for 1 year after receipt. After reconstitution, store at 2~8°C, valid for 7 days. After reconstitution, store at -85~-65°C, valid for 3 months.
Reconstitution Method
Before opening the vial, centrifuge first to bring the contents to the bottom. Reconstitute with sterile distilled water or PBS to a concentration of 0.1-1.0 mg/mL. It can be further diluted according to subsequent experiments. For long-term storage, it is recommended to add carrier protein (0.1% BSA) to the dilution buffer. Aliquot into single-use amounts, store at -80°C, and avoid repeated freeze-thaw cycles.
