The Human Organoid Cytokine Kit contains four cytokines: Wnt3a, R-spondin-1, Noggin, and EGF. These four cytokines are key components for intestinal organoid culture.

Wnt-3a is one of the most critical factors. The Wnt signaling pathway is the main driver for the maintenance and self-renewal of intestinal stem cells. It activates β-catenin signaling, driving cell proliferation and inhibiting differentiation. In organoid culture, mimicking the Wnt signal provided by Paneth cells at the base of the crypt is essential for establishing and expanding organoids. Colon organoids are generally more dependent on Wnt.

R-spondin-1 is a potent enhancer of Wnt signaling. It amplifies and stabilizes Wnt signals by binding to the LGR5 receptor (a marker of intestinal stem cells) and inhibiting endogenous inhibitors of the Wnt signaling pathway. RSPO1 acts synergistically with Wnt-3a to maximize Wnt signal activity, thereby efficiently maintaining the LGR5+ stem cell population. Without R-spondin, organoids are difficult to survive long-term.

Noggin is an inhibitor of the Bone Morphogenetic Protein (BMP) signaling pathway. BMP promotes cell differentiation; inhibiting it is necessary to maintain the stem cell state and help maintain crypt-like structures.

EGF stimulates epithelial cell proliferation and migration. It provides a basic pro-mitogenic signal to support the continuous growth and expansion of organoid cells.
