The sequence of Hieff™ Unmethylated Lambda DNA is completely unmethylated, which can be used as a known "negative" or "0% methylation" standard to evaluate the efficiency and accuracy of the experimental workflow. After sequencing, theoretically all C should be converted to T. By calculating how many C have not been converted (i.e., still read as C), the bisulfite conversion efficiency of the experiment can be precisely evaluated, ensuring the reliability of subsequent analysis. The full length of the Lambda DNA sequence is 48.5 kb.

Notes

1. As a "spike-in" (negative control), it is recommended to spike into real samples at a ratio of 0.1%–1% and proceed with the entire experimental workflow together.

2. The concentration of Unmethylated Lambda DNA is 2 ng/μL, and the total amount of Unmethylated Lambda DNA is 1 μg.
