Product introduction

The Human T-Cell Activation Kit is a standardized reagent set designed to mimic the “dual signals” provided by antigen-presenting cells (APCs) in vivo, enabling efficient activation, stable expansion, and functional maintenance of human naïve/ resting T cells. It can be directly applied to PBMCs or purified CD3+ T cells and is widely suitable for basic immunology research as well as the development of cell therapies such as CAR-T.

Activation Principle (Dual-Signal Mimicry):

First Signal (Antigen Recognition Signal): An anti-human CD3 antibody binds to the CD3/TCR complex on the surface of T cells, triggering the core signaling pathway essential for T-cell activation—effectively replacing the stimulation provided by the antigen-MHC complex.

Second Signal (Co-stimulatory Signal): An anti-human CD28 antibody binds to CD28 on the surface of T cells, delivering the critical co-stimulatory signal necessary to prevent T-cell anergy and significantly enhance proliferation and survival.

Growth Signal (Expansion and Maintenance): Recombinant human IL-2 promotes clonal expansion of activated T cells, maintaining their activity and functionality—including effector functions such as CD8+ cytotoxicity and CD4+ helper activity.

Storage Conditions

Store at -25°C to -15°C. Once received, the product has a shelf life of 1 year. Avoid repeated freeze-thaw cycles; before opening the vial, centrifuge it first. It is recommended to aliquot the product into single-use portions to prevent repeated freeze-thaw cycles.

Human IL-2 Reconstitution Method

Before opening the vial, first centrifuge it to bring the contents down to the bottom. Resuspend the lyophilized material using sterile distilled water or PBS, and reconstitute it to a concentration of 0.1–1.0 mg/mL. Further dilution can be performed as needed for subsequent experiments. For long-term storage, it is recommended to add a carrier protein (0.1% BSA) to the dilution buffer. Aliquot the solution into single-use amounts, and store at -80°C, avoiding repeated freeze-thaw cycles.
