Product introduction
The human breast organoid culture cytokine kit contains seven key factors: Wnt3a, R-spondin-1, Noggin, EGF, FGF-10, NRG1, and bFGF. Here is a breakdown of their specific roles:
Wnt3a is one of the most critical factors and serves as a key switch for maintaining stemness. As a ligand for the Wnt signaling pathway, it activates β-catenin signaling, directly driving the self-renewal and proliferation of breast stem/progenitor cells.
R-spondin-1 acts as a potent enhancer of Wnt signaling. While it does not directly activate the Wnt pathway itself, it binds to the Lgr5 receptor to inhibit Wnt-suppressing proteins (RNF43/ZNRF3), thereby dramatically amplifying the signal strength of Wnt3a.
Noggin functions as a differentiation inhibitor. As an antagonist of Bone Morphogenetic Proteins (BMPs), it counteracts BMP signaling, which typically induces cell differentiation (particularly toward a mesenchymal fate or halts proliferation). By inhibiting this pathway, Noggin ensures that cells remain in an undifferentiated stem cell state.
EGF provides the fundamental drive for proliferation. By binding to cell surface EGFR, it activates downstream pathways such as MAPK/ERK, promoting mitosis and survival in epithelial cells.
FGF-10 mimics epithelial-mesenchymal signaling. In vivo, FGF-10 is primarily secreted by mesenchymal cells to act on epithelial cells. In culture, it promotes the proliferation, migration, and branching morphogenesis of breast epithelial cells.
NRG1 (Neuregulin 1) is essential for luminal cell differentiation and expansion. As a ligand for ErbB receptors (such as HER3/HER4), NRG1 signaling is crucial for the survival and proliferation of luminal epithelial cells in the breast.
Prolactin activates the JAK2/STAT5 pathway, prompting organoids to express milk proteins (such as β-casein) and secrete lipid droplets, effectively simulating the functional state of the lactating breast.

Storage Conditions
Store at -85℃ ~ -65℃. Shelf life is 1 year from the date of receipt. Avoid repeated freeze-thaw cycles; before opening the vial, centrifuge it first. It is recommended to aliquot the product into single-use portions for each experiment and then store at -80℃ after aliquoting.
Reconstitution instructions
Before opening the vial, first centrifuge it to bring the contents down to the bottom. Resuspend the material using sterile distilled water or PBS, and reconstitute it to a concentration of 0.1–1.0 mg/mL. Further dilution can be performed as needed for subsequent experiments. For long-term storage, it is recommended to add a carrier protein (0.1% BSA) to the dilution buffer. Aliquot the solution into single-use amounts, and store at -80°C, avoiding repeated freeze-thaw cycles.
