The Hifair™ CoAmp Multiplex One Step RT-qPCR Probe Kit (UDG Plus) is a multiplex one-step fluorescent quantitative PCR master mix designed for the efficient simultaneous detection of DNA and RNA targets within the same reaction system. By carefully integrating high-performance thermostable reverse transcriptase and hot-start DNA polymerase, along with a proprietary reaction buffer system, this kit achieves excellent amplification efficiency and balanced detection performance for both DNA and RNA templates. It effectively overcomes the amplification bias issues commonly encountered with traditional reagents during DNA/RNA co-detection.
The core components of the kit include the deeply optimized 5× Hifair™ MP Buffer and a master mix enzyme system. The buffer contains sufficient Mg²⁺, dNTPs, and other essential components for amplification, and incorporates specific amplification enhancers and multiplex reaction stabilizers. This ensures high amplification efficiency while supporting robust multiplex detection of up to four targets. Additionally, the system incorporates dUTP and Uracil-DNA Glycosylase (UDG), which efficiently degrades carryover PCR products, fundamentally preventing amplification product contamination and ensuring the reliability and reproducibility of experimental results.

Using Reagent 16929, the simultaneous detection of African Swine Fever Virus (DNA) and Porcine Reproductive and Respiratory Syndrome Virus (RNA) was performed in a single reaction well. Results demonstrate that both targets achieved amplification efficiencies exceeding 90%, with no amplification bias, ensuring balanced and reliable outcomes.

(A) Accelerated Stability: After incubation at 37°C for 14 days, Reagent 16929 showed no changes in detection rate, Ct values, or peak shape compared to the -20°C control, demonstrating excellent thermal stability.
(B) Freeze-Thaw Stability: The reagent maintained consistent amplification performance for both DNA-HPIV and RNA-HCOV after 50 dry ice freeze-thaw cycles.
