Proteinase K is a broad-specificity serine protease with a relative molecular weight of approximately 29.3 kDa. It cleaves peptide bonds at the carboxyl side of aliphatic and aromatic amino acids. Proteinase K has wide applications, including preparation of high-molecular-weight chromosomal DNA for pulsed-field gel electrophoresis, Western blotting (protein immunoblotting), and removal of nucleases during DNA and RNA preparations. Denaturants such as SDS (1%) can enhance its activity. The typical working concentration is 50–100 µg/mL, with the exact concentration determined based on factors such as the presence of SDS or urea in the buffer, pH, and temperature.
This product is a low-residual Proteinase K, which maintains the enzyme's inherent properties while featuring significantly reduced background microbial residues, ensuring greater accuracy in downstream detection applications.

- Derived from Tritirachium album (also known as Parengyodontium album or Limber strain in some references);
- Specific activity: ≥800 U/mL;
- Protein concentration: ≥20 mg/mL;
- Free of RNase and DNase contamination;
- Stable activity in the presence of urea and SDS;
- Active over a broad pH range (pH 4.5–12.5), with optimal activity between pH 7.0–12.0;
- Reaction conditions: Incubate in reaction buffer at 37°C–65°C for 1 hour. Recommended temperature: 65°C;
- Unit definition: One unit is the amount of enzyme required to hydrolyze casein to release 1 µmol of L-tyrosine per minute under defined conditions;
- Inactivation method: Add PMSF or DFIP inhibitor to the reaction system for complete inactivation.
