Agarose is a purified linear galactoglycan hydrophilic colloid derived from agar or agar-containing algae. It is a linear polymer composed of β-d-galactosyl (1-4) linked to 3, 6-dehydrated-L-galactosyl. As a gel reagent, it is commonly used for routine nucleic acid analysis by gel electrophoresis or blotting (such as Northern or Southern) and for protein applications, such as radiative immunodiffusion (RID) assay.

Q: Is the 10208ES Low Melting Point Agarose sterile and nuclease-free? What about other types of agarose?
A: It is sterile and nuclease-free. Low melting point agarose is commonly used for plaque assays, so it can be used safely for cell culture. Other types of agarose are also sterile and nuclease-free.
Q: Why is the 10208ES agarose relatively soft? What is the reason for this?
A: The firmness is related to the gel strength. Agarose is extracted from agar, and there will always be some variation in strength levels; it is impossible for every batch to be identical. The gel strength specification for this agarose is $\ge 1200$, and as long as it is above this range, it is considered qualified.
Q: Why do the bands appear smeared when using 10208ES?
A: The gelling time may have been too short, resulting in an uneven gel. For a 1% agarose gel, the minimum gelling time is 30 minutes.
Q: What size range of bands can high-resolution agarose separate?
A: The resolution for both high-resolution agarose and 3:1 agarose is around 2% (meaning if the fragment size is 30 bp, it can separate $30 \times 0.02$; for a 100 bp fragment, $100 \times 0.02$).
Q: Can 10208ES low melting point agarose be used for pulsed-field gel electrophoresis (PFGE)?
A: No, pulsed-field gel electrophoresis requires very low electroosmosis and very high gel strength.
