[Whole Transcriptome Sequencing - Research on the Targeted Regulation and Interaction Relationship between Non-coding RNA and mRNA]
Whole transcriptome high-throughput sequencing adopts the ribosome-depleted strand-specific library construction method and the small fragment enrichment screening library construction method, which can achieve the library construction, sequencing, information analysis ,joint analysis and ceRNA, etc. of coding RNA and non-coding RNA, So in that way it can obtain the whole RNA transcript data information related to specific biological processes (such as development, disease, etc.) quickly, comprehensively and accurately. Through data analysis, the research on biological regulation mechanism can be extended to a three-dimensional model combining "network and multi-level", which is helpful to comprehensively interpret biological phenomena.
There are two ways of whole transcriptome sequencing, one is LncRNA-Seq + Small RNA, and the other is LncRNA-Seq + Small RNA + CircRNA, which can obtain more comprehensive information of CircRNA.

2.Whole transcriptome sequencing with three libraries: LncRNA-Seq + Small RNA + CircRNA

LncRNA Research
Long non-coding RNA (LncRNA) is a kind of non-coding RNA with unique regulatory function that has attracted extensive attention in recent years. The length of LncRNA is more than 200nt, it does not encode proteins, and its conservation among species is poor. Some of its sequence structures are similar to mRNA (containing polyA tail and alternative splicing), while others do not have these characteristics. At present, the generation and functional mechanism of LncRNA are not yet fully understood, however, many LncRNAs with important biological functions (such as cancer occurrence, X chromosome silencing and the formation of complex regulatory networks with other regulatory factors) have been discovered.

Eukaryotic Transcriptome Research
Eukaryotic transcriptome research uses RNA-seq technology to obtain the total mRNA that can be transcribed by specific cells in a certain functional state. Then, the downloaded raw data is spliced and assembled, the gene expression level is counted, and differential analysis and functional enrichment analysis are carried out. It can not only obtain most of the gene information of the species, but also study the gene expression difference and functional pathway change at the overall level, and reveal the molecular mechanism of specific biological processes.
