Endonuclease IV (also known as Nfo) is an endonuclease whose main function is to recognize apurinic/apyrimidinic (AP) sites in DNA strands. It cleaves the phosphodiester bond at the 5′ side of the AP site, generating a 3′-hydroxyl group and a 5′-deoxyribose phosphate terminus (dRP). In addition, this enzyme also has 3′-phosphodiesterase activity and can release glyceraldehyde-3-phosphate, intact deoxyribose 5′-phosphate, and inorganic phosphate from the 3′ end of DNA.

Case Study 1: No Non-specific DNase Residue
In a 20 μL reaction system, 100 U of Endonuclease IV from different batches and 0.5 μg of λDNA-HindIII digest were added and incubated at 37°C for 16 h. The DNA band pattern did not change in agarose gel electrophoresis, indicating that Yeasen Endonuclease IV contains no non-specific DNase residue.

Case Study 2: Host (E. coli) gDNA Residue < 0.05 copies/10 U
Host (E. coli) genomic DNA residue was tested for different batches of Yeasen Endonuclease IV. The results show that the host gDNA residue of Yeasen Endonuclease IV is < 0.05 copies/10 U.
