Creatine kinase (CK) is mainly found in the cytoplasm and mitochondria. It is an important kinase directly related to intracellular energy metabolism, muscle contraction, and ATP regeneration. CK mainly catalyzes the conversion of adenosine diphosphate (ADP) and phosphocreatine to ATP and creatine, playing a critical role in rapid ATP regeneration in cells with high energy demand. In RPA technology, ATP generated by creatine kinase-catalyzed hydrolysis supplies energy, thereby altering the conformation of the recombinase (T4 UvsX)-primer complex, dissociating the recombinase to expose the 3′ end of the primer, and thus promoting the binding of strand-displacement Bsu DNA polymerase to the primer to initiate DNA amplification.
