Reverse transcription is the process of converting RNA into complementary DNA (cDNA), which is mediated by the reverse transcriptase enzyme. In the presence of annealing primers, the reverse transcriptase first binds to the RNA template. The RNA-dependent DNA polymerase activity of the reverse transcriptase synthesizes cDNA by incorporating dNTPs from the reaction mixture. This process results in the formation of a cDNA:RNA hybrid strand, which is known as the first-strand cDNA synthesis.

The Moloney Murine Leukemia Virus (M-MLV) reverse transcriptase is one of the commonly used reverse transcriptases in molecular biology laboratory procedures. To achieve higher reverse transcription efficiency, better thermal stability, and an increased rate of cDNA synthesis, the original reverse transcriptase is often genetically engineered. Currently, the reverse transcription reagents available on the market typically require reaction times ranging from 15 to 30 minutes, which is relatively long.
